    Antioxidant and antihyperglycemic activities of Scorzonera cinerea radical leaves in streptozocin-induced diabetic rats

    Scorzonera species are used for treating various diseases. They are consumed raw, especially in the spring, and have nutritious and dietetic values. This study evaluated the antidiabetic and antioxidant effects of ethanolic extracts of Scorzonera cinerea (Sc) radical leaves in diabetes mellitus. Five random groups of Wistar rats (n = 8) were created - control, diabetic, acarbose, Sc-Dried, and Sc-Frozen. Phenolic profiles of extracts were determined by HPLC. Free radical scavenging capacity was measured using DPPH and ABTS tests. The inhibitory effects of Sc extracts on α-glucosidase and α-amylase activities were also evaluated. Moreover, superoxide dismutase (SOD), glutathione peroxidase (GPx), and catalase (CAT) activities, glutathione (GSH) concentration, malondialdehyde (MDA), total antioxidant status (TAS) and total oxidant status (TOS) were analyzed in theliver tissues. While dried Scorzonera extract was more effective in α-amylase inhibitory activity, frozen Scorzonera extract was more effective in α-glucosidase inhibitory activity. Sc-Dried and Sc-Frozen extracts lowered blood glucose and HbA1c levels, they also increased insulin. Although liver MDA and TOS were significantly increased in the diabetic group, their values were significantly lower in the Sc-Dried- and Sc-Frozen-treated groups. GSH, TAS, and anti-oxidant enzyme activities decreased in the diabetic group, but Sc-Dried and Sc-Frozen supplements significantly enhanced liver antioxidant values. In conclusion, S. cinerea treatment exerts potential hypoglycemic and antioxidant effects in diabetes. Thus, it can be considered as a candidate dietary supplement for health benefits in diabetes.

    Antioxidant and hepatoprotective effect of oriental hackberry (celtis tournefortii l.) against CCI4 injury in rat liver

    This study aimed to investigate the antioxidant and hepatoprotective effects of Celtis tournefortii leaf extract (Ct) against CCl4-induced liver damage in rats. A total of 32 Wistar albino rats were divided into four equal groups (n = 8): control, CCl4 group, CCl4 + Celtis group (CCl4 + Ct), and Celtis group (Ct). Superoxide dismutase (SOD), glutathione peroxidase (GPx), and catalase (CAT) activities, glutathione (GSH) level, malondialdehyde (MDA) content, total antioxidant status (TAS), and total oxidant status (TOS) were analyzed in the liver tissues. Liver enzymes were assayed in the serum. The liver histopathology was also evaluated. The levels of alanine aminotransferase and lactate dehydrogenase significantly decreased in the CCl4 + Ct group in comparison with those in the CCl4 group. SOD and GPx activities and TAS increased significantly with CCl4 + Ct administration when compared with the administration of CCl4 alone. The oxidative stress parameters, such as MDA and TOS, showed a reduction when Ct treatment was applied to the CCl4-induced group. The results revealed that Celtis remarkably attenuated the degenerative and necrotic changes caused by CCl4 in the liver tissues. Consequently, the Ct may augment the antioxidant status and moderated the severity of CCl4-induced liver damage. The therapeutic use of Celtis tournefortii might be effective in the treatment of liver injuries.

    The effect of bioactive compounds of prunus spinosa leaves and flowers mixture on the antioxidant capacity in streptozotocin-induced diabetic rats

    Diabetes is a serious and chronic public health problem worldwide. Hyper-glycemia leads to the production of free radicals thus impairing oxidant/anti-oxidant balance and causing various complications in diabetes. Herbs can pro-vide natural remedies against various diseases in traditional medicine. Phyto-chemicals especially phenolic compounds have some biological properties such as antioxidant and antidiabetic effects. Plant parts of Prunus spinosa are used in traditional medicine against various disorders. This study evaluated the mixture of Prunus spinosa leaves and flowers extract (PSE) on the antioxidant capacity in diabetes. The phenolic profile of PSE was determined by HPLC method. Antioxidant capacity was evaluated with radical scavenging activities as well as total phenolic and flavonoid content. Seven random groups (n = 8) of rats were constituted to evaluate the antioxidant capacity - control, dia-betic, PSE25, PSE50, insulin, metformin, and acarbose. Total oxidant status (TOS) and total antioxidant status (TAS) were analyzed in the liver tissues. Chlorogenic acid and ellagic acid were found to be the main compound in the mixture of P. spinosa leaves and flowers extract. Antioxidant capacity of PSE exhibited effective radical scavenging activity. Although oxidative stress seri-ously increased in diabetes, PSE25 and PSE50 supplementation significantly recuperated liver antioxidant status. In conclusion, PSE markedly has antioxi-dant capacity through its high phenolic compounds, thus PSE treatment may have potential antioxidant effects for health benefits in diabetes.

    The effect of olive leaf extract on digestive enzyme inhibition and insulin production in streptozotocin-induced diabetic rats

    Olive leaf has natural bioactive compounds, mainly oleuropein, that are widely considered to have potentially beneficial effects on health. This study aimed to evaluate the effects of olive leaf extract (OLE) on the inhibition of carbohydrate digestive enzymes, and immunohistochemical study of insulin in the pancreas of in vivo streptozotocin-induced diabetic rats. Blood glucose levels, insulin, glycated hemoglobin (HbA(1c)), alpha-amylase and alpha-glucosidase activities, and an immunohistochemical study were performed at the end of the experiment. In the OLE treated group, blood glucose levels and HbA(1c) significantly decreased while insulin levels increased. Besides this, OLE treated group showed remarkable inhibitory activities on alpha-amylase and alpha-glucosidase compared with the Acarbose treated group. It was observed that OLE exhibited partial positive immunoreaction for insulin in beta-cells through immunohistochemical analysis. Considering that OLE is more tolerable for digestion system compared to acarbose, it may be a better fitoformulation for antidiabetic medications. OLE could offer an additional beneficial effect for the treatment of diabetes.

    The effects of melatonin and Vitamin E on antioxidant parameters in copper nephrotoxicity

    Copper (Cu) compounds have been widely used in various fields especially industrial processes and agriculture. Although trace amounts of Cu are essential to sustain life, excessive Cu is toxic for living organisms. This study aims to investigate the effects of vitamin E and melatonin on antioxidant enzymes activities in kidney against acute Cu toxicity. In this study, 28 Wistar albino rats were divided four equal groups (n=7) as control, Cu, Cu+Vitamin E, and Cu+Melatonin groups. Superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase (CAT) enzymes activities and protein carbonyl content were analyzed. Also, kidney function tests were evaluated. While Cu administration increased creatinine, blood urea nitrogen (BUN), uric acid, Na, K and Mg levels, vitamin E and melatonin was effective reduction on these levels. Carbonyl level significantly decreased in Cu+Vitamin E and Cu+melatonin groups compared to Cu group. Although SOD, GPx and CAT activities in tissue decreased by Cu exposure, vitamin E and melatonin treatment showed significant increase on their activities. Consequently, vitamin E and melatonin supplements may provide prevention Cu -based oxidation by strengthening the antioxidant defense system. In this respect, it may show a protective effect by preventing the development of nephrotoxicity.

    Effects of scorzonera cinerea on immune system and hematological parameters in short-term hyperglycemia

    Medicinal herbs offer natural remedies for various ailments because of their comprehensive effects. Scorzonera cinerea L. (Sc) is an edible wild plant and is used in traditional medicine against various diseases. The purpose of this study was to investigate the immune system effects of S. cinerea radical leaf extract in diabetic rats.Methods: The 5 groups were formed - Control, Diabetic, Sc-Dried, Sc-Frozen, and Acarbose. Adenosine deaminase (ADA), xanthine oxidase (XO), and myeloperoxidase (MPO) activities in liver and kidney tissues were analyzed. Hematological parameters were also evaluated.Results: ADA, XO, and MPO activities in both tissues significantly increased in diabetic conditions. However, Sc treatments significantly decreased liver ADA, liver and kidney XO, and liver and kidney MPO activities. There was no significant change in red blood cell (RBC) parameters. Although platelet (PLT) count and MPV were raised in the diabetic group, these parameters were reduced with Sc-Dried and Sc-Frozen treatment. While lymphocyte (LYM%) significantly decreased in defense cells in the diabetic group, neutrophil (NEU%), monocyte (MO%), and eosinophil (EOS%) elevated. However, a significant decrease was observed in NEU% and EOS% with Sc treatment.Conclusion: S. cinerea treatment can exert a potential immunoregulatory effect in diabetes. Thus, S. cinerea can be considered as an adjuvant to augment immune system on diabetes.

    Hazelnut by-products as a valuable resource: Lipid peroxidation inhibition effect, bioaccessibility and antidiabetic properties

    The aim of this study is to determine the chemical and molecular properties of hazelnut by-products, to measure the bioaccessibility values of phenolic compounds found in the structure of these wastes and to reveal their antidiabetic and lipid peroxidation (LPO) inhibition effects. The highest moisture and protein content were found in skin samples with 6.33% and 7.70%, respectively. The predominant fatty acid in hazelnut skin is oleic acid, which constitutes 78.65% of the total fatty acid composition. The highest antioxidant activity value was measured in the shell with an IC50 value of 19.08 mg/mL. The sample with the highest content of gallic acid and protocatechuic acid content was the skin extract. The dominant phenolic component in the shell and husk samples was kaempferol, while in the skin sample it was rutin. Shell bioaccessibility was found to be much higher for o-coumaric acid (45.83%), rutin (49.25%) and kaempferol (46.32%) components. The bioaccessibility of the shell sample in total phenolic concentration was measured as 45.87%. The highest LPO inhibition value belonged to the shell sample with an IC50 of 4.50 mg/mL, while the lowest value belonged to the husk sample with an IC50 value of 25.03 mg/mL. The highest antidiabetic effect was observed in the shell sample with values of 3.55 mg/mL and 25.44 mg/mL for α-amylase and α-glucosidase inhibition, respectively. The highest recovery belonged to 4-Hydroxybenzoic acid in the skin sample. As a result, it was concluded that hazelnut by-products, especially the shell, exhibit high antioxidant effects, have antidiabetic and LPO inhibition properties thanks to their bioactive components and high bioaccessibility values. Thus, they have valuable usage opportunities in the fields of food, pharmacology and medicine. © 2025 Elsevier Ltd

    Investigation of phenolic composition, antioxidant capacity, and antidiabetic effect of ornithogalum Lanceolatum L.: An in vitro Study

    Diabetes Mellitus is a global health problem that leads to macro- and microvascular diseases associated with hyperglycemia. Phytotherapy is one of the alternative ways to cope with this type of disease. The genus Ornithogalum is consumed as a wild edible plant and traditionally used for ailments. This study aims to investigate the phenolic composition using High-Performance Liquid Chromatography as well as antioxidant and antidiabetic effects using spectrophotometric method of Ornithogalum lanceolatum L. aerial parts and bulb. In order to determine the antioxidant capacity total phenolic content, total flavonoid content and DPPH and ABTS free radical scavenging activities were analyzed in O. lanceolatum. Moreover, in vitro inhibitory effects of the O. lanceolatum aerial parts and bulb on digestive enzymes were determined by evaluating the α-amylase and α-glucosidase activities. Protocatechuic acid was found to be the main compound in both plant parts. However, the amounts of the total phenolic acids and flavonoids were found higher in the aerial parts than those in bulb as well. Furthermore, O. lanceolatum aerial parts exhibited more radical scavenging activity than bulb. The α-amylase and α-glucosidase IC50 inhibition activities of aerial parts were found more efficient than those for bulb. It can be concluded that O. lanceolatum can enhance the antioxidant status and also can prevent nutraceutically postprandial hyperglycemia by inhibiting α-amylase and α-glucosidase enzymes. These findings reveal the importance of traditional remedies in the ethnopharmacological use of herbs.

    Mixture of leaf and flower extract of Prunus spinosa L. Alleviates hyperglycemia and oxidative stress in streptozotocin-induced diabetic rats

    Diabetes Mellitus is a global health problem that leads to various complications associated with hyperglycemia. In traditional medicine, herbal treatment is one of the alternative ways to cope with this type of disease. The aim of this study is to investigate the antidiabetic and hepato-pancreatic protective effects of the mixture of Prunus spinosa leaves and flowers (PSE) extract in streptozocin-induced diabetes mellitus. Seven random experimental groups of Wistar rats (n = 8) were created as followed; control, diabetic, PSE25, PSE50, insulin, metformin, and acarbose. α-amylase and α-glucosidase inhibitory activities of PSE were determined. Antioxidant enzymes activities and lipid peroxidation were analyzed in the liver tissue. Histopathological examination of liver and pancreas was also performed. α-amylase and α-glucosidase IC50 inhibition values of PSE were found more efficient, comparing to those of standard acarbose. While blood glucose levels severely increased in all diabetic groups, PSE25 and PSE50 treatments were effective in regulating blood glucose levels. Moreover, administration of PSE25 and PSE50 improved insulin levels compared to the diabetic group. Although increased oxidative stress in the diabetes seriously suppressed antioxidant activities, PSE25 and PSE50 supplementation significantly recuperated liver antioxidant capacity. Despite severe degenerative and necrotic changes in diabetes, these findings alleviated with PSE administrations. Moreover, PSE treatments remarkably recuperated β-cells. These results reveal that there may be an alternative way to control high blood glucose levels, which is one of the most important complications of diabetes. Furthermore, PSE can provide a protection against oxidative stress, liver and pancreatic damage by augmenting antioxidant capacity in diabetes.

    Modulation of the Immune System and Hemogram Parameters by Prunus spinosa in Short-Term Hyperglycemia

    Diabetes mellitus (DM) is one of the chronic diseases, relationship increased blood glucose level, that requires urgent global attention due to its prevalence and associated complications. DM leads to oxidative stress that plays an important role in the development of various complications in diabetes by suppressing the immune system. Prunus spinosa is a plant that has been used in the treatment of many diseases from past to present, thanks to its high antioxidant activity. Therefore, the present study aims to research the effect of P. spinosa leaf and flower mixture on immune system during the short-term diabetic condition. In the study, 56 Wistar albino male rats divided into 7 groups, one of which control and others six diabetic groups, were used to determine the effects of P. spinosa on adenosine deaminase (ADA), (xanthine oxidase) XO and myeloperoxidase (MPO) activities in the liver tissues of diabetic rats as well as on hemogram parameters. Two of these groups were given plant extract in different concentrations (25 and 50 mg/kg bw) and the results were compared with insulin, metformin and acarbose groups. The results showed that both doses administered had a modulating effect on the changing hematological parameters caused by diabetes. Treatment groups significantly decreased ADA, XO, and MPO activities compared to diabetic group. The effects of the PSE50 were found to be more effective than all other treatment. These effects of the plant in diabetesmay be due to its therapeutic immunoregulatory potential. As a result, P. spinosa can be a valuable resource as an adjuvant on diabetes.

    Protective effect of celtis tournefortii against copper-induced toxicity in rat liver

    This study aimed to investigate the antioxidant and hepatoprotective effects of Celtis tournefortii fruit extract (Ct) against copper-induced liver damage in rats. Thirty-two Wistar Albino rats were divided into four equal groups (n = 8): Control, Copper (Cu), Copper + C. tournefortii (Cu+Ct), and C. tournefortii (Ct). Superoxide dismutase (SOD), glutathione peroxidase (GPx), and catalase (CAT) activities, glutathione (GSH) concentration, malondialdehyde (MDA), total antioxidant status (TAS) and total oxidant status (TOS) were analysed in the liver tissues. Liver histopathology was also evaluated. Alanine aminotransferase and lactate dehydrogenase significantly decreased in the Cu+Ct group compared with the Cu group. Oxidative stress parameters MDA and TOS significantly increased with copper administration, whereas they decreased with C. tournefortii co-treatment compared to Cu group. GSH concentration and TAS showed significant decreases with copper administration. Celtis tournefortii co-supplementation with copper significantly enhanced antioxidant parameters such as TAS, SOD, and GPx. Celtis tournefortii remarkably attenuated degenerative and necrotic changes caused by the oral exposure of copper in the liver tissue of rats. Celtis tournefortii may provide amelioration of the antioxidant status and moderation of severity of liver damage against copper toxicity. The therapeutic use of C. tournefortii may exhibit protective effects in liver injury treatment.

    Sisymbrium altissimum yapraklarının fenolik bileşen, antioksidan kapasite ve antidiyabetik özelliklerinin tespiti

    Diabetes Mellitus (Şeker hastalığı), yüksek kan glukozu (hiperglisemi) ile karakterize edilen ve tüm dünyada en çok muzdarip olunan hastalıklar arasındadır. Hiperglisemi oksidatif stresin oluşum mekanizmalarını tetikleyerek pek çok hastalığın patogenezinde rol oynar. Fitoterapi bu hastalıklar ile başa çıkmada alternatif yollardan biridir. Tıbbi, aromatik ve yabani yenilebilir bitkiler içerdikleri fitokimyasallar sayesinde nutrasötik olarak çeşitli hastalıklara karşı koruyucu etki göstermelerinin yanı sıra besleyici ve diyetetik özelliklere sahiptirler. Sisymbrium türleri yabani yenilebilir bitkiler arasında olup geleneksel olarak çeşitli hastalıkların tedavisinde kullanılmaktadır. Bu çalışmada, Sisymbrium altissimum yapraklarının fenolik bileşiminin yanı sıra antioksidan kapasitesinin ve antidiyabetik özelliklerinin araştırılması amaçlanmıştır. Bu maksatla S. altissimum yaprak ekstraktında (SAE) yüksek performanslı sıvı kromatografisi (HPLC) ile fenolik bileşen tespiti yapıldı. Ayrıca, antioksidan kapasitesini belirlemek amacıyla toplam fenolik ve toplam flavonoid madde tayini ile 2,2-difenil-1-pikrilhidrazil (DPPH) ve 2,2′-Azino-bis (3-etilbenzotiazolin-6-sülfonik asit) (ABTS) serbest radikal temizleme aktivite analizleri gerçekleştirildi. Yaprakların antidiyabetik etkisi, α-amilaz ve α-glukozidaz inhibisyon aktivite tayini ile ölçüldü. Elde edilen sonuçlarda, SAE’nin önemli düzeyde antioksidan aktivite gösterdiği belirlendi. Yapraklarda en çok bulunan fenolik bileşenin rutin olduğu ayrıca, kuersetin ve protokateşuik asidin ise yapraklardaki diğer baskın fenolik bileşenler olduğu saptandı. Diğer taraftan yaprakların α-amilaz ve α-glukozidaz inhibisyon aktivitelerinin akarboz ile karşılaştırıldığında daha düşük olduğu tespit edildi. Sonuç olarak, S. altissimum’um önemli fenolik bileşen içeriğiyle antioksidan durumu güçlendirebileceği ve ayrıca kısmi α-amilaz ve α-glukozidaz inhibisyon aktiviteleri sayesinde nutrasötik olarak yemek sonrası hiperglisemiyi önleyebileceği sonucuna varılabilir.

    Zeytinde bulunan polifenoller ve farmakolojik etkileri

    Akdeniz tarzı beslenmenin vazgeçilmezi olan zeytinin sunmuş olduğu faydaların başında antioksidan etkisi vardır. İçerdiği zengin fenolik bileşikler sayesinde oksidatif stresin azaltılmasında önemli bir rol oynamaktadır. Bu sayede diğer rahatsızlıkların ve hastalıkların önlenmesinde ya da tedavisinde ayrıca fayda sağladığı söylenebilir. Zeytin ve yaprağından elde edilen ektstraktının yanın da içerdiği önemli fenolik bileşikler arasında olan Oleuropein ve Hidroksitirozol pek çok çalışmaya konu olmuştur. Bunlar arasında en çok antioksidan, antidiyabetik, antimikrobiyal, anti-inflamatuar, antihipertansif ve antikanser etkiler araştırılmıştır. Zeytin zengin biyoaktif bileşikleri ile hem bireysel hem de sinerjistik olarak çeşitli farmakolojik etkiler göstermektedir. Zeytinde en bol bulunan ve en aktif madde olan Oleuropeinin enzimlerin inhibe, modüle ve/veya regüle edilmesinde baş rolü oynadığı söylenebilir.
